目的

探讨1型糖尿病(T1D)患者血清髓源性免疫抑制细胞(MDSCs)与免疫球蛋白G4(IgG4)的关系。

方法

选取汕头大学医学院第二附属医院于2015年1月至2017年1月期间收治的T1D患者30例,2型糖尿病(T2D)患者20例;另选取同一家医院2015年1月至2017年1月期间健康体检者20例作为对照组。采用酶联免疫吸附法测定血清IgG4浓度,流式细胞分析术检测外周血MDSCs表达频率。采用t检验以及Mann-Whitney U检验比较3组间血清IgG4浓度及MDSCs表达频率的差异,采用Spearman相关分析法分析血清IgG4与MDSCs的相关性。

结果

T1D组血清IgG4为(0.92±0.79)g/L,T2D组血清IgG4为(0.54±0.33)g/L,对照组血清IgG4为(0.34±0.15)g/L,T1D血清IgG4浓度高于对照组,差异具有统计学意义(t=4.900,P<0.001),T1D组高于T2D组,差异具有统计学意义(t=2.830,P=0.007),T2D组高于对照组,差异具有统计学意义(t=2.466,P=0.010)。T1D组外周血MDSCs表达频率为(4.09±1.53)%,T2D组外周血MDSCs表达频率为(2.86±1.63)%,对照组外周血MDSCs的表达频率为(1.93±0.76)%。T1D组外周血MDSCs的表达频率明显高于对照组,差异有统计学意义(Z=-9.650,P<0.01)。T1D组MDSCs表达频率高于T2D组,差异有统计学意义(t=2.899,P=0.005)。血清IgG4与MDSCs呈正相关(r=0.482,P<0.05)。

结论

T1D患者MDSCs与IgG4升高,且IgG4与MDSCs呈正相关,具有重要研究意义。

Objective

To assess the relationship between serum myeloid-derived suppressor cells (MDSCs) and immunoglobulin G4 (IgG4) in patients with type 1 diabetes mellitus (T1DM).

Methods

Thirty patients with T1DM and 20 patients with T2DM were selected at the Second Affiliated Hospital of Shantou University Medical College from January 2015 to January 2017; another 20 healthy subjects were selected from January 2015 to January 2017 as a control group. The frequency of MDSCs in peripheral blood was detected by enzyme-linked immunosorbent assay and flow cytometry. The t-test, chi-square test, Mann-Whitney U test and spearman correlation analysis were used to compare the changes of serum IgG4 content, MDSCs frequency and the correlation between serum IgG4 and MDSCs.

Results

Serum IgG4 level was (0.92±0.79) g/L in the T1DM group, (0.54±0.33) g/L in the T2DM group, and (0.34±0.15) g/L in the normal control group. Compared with the control group, serum IgG4 levels in the T1DM and T2DM groups were significantly higher (t=4.9, P<0.001; t=2.466, P=0.01). Compared with the T2DM group, serum IgG4 level in the T1DM group was significantly higher (t=2.83, P=0.007). The frequency of MDSCs in the T1DM group was significantly higher than those in the control group (Z=-9.650, P<0.01) and in the T2DM group (t=2.899, P=0.005). Serum IgG4 levels in T1DM patients were positively correlated with the frequency of MDSCs (r=0.482, P<0.05).

Conclusion

MDSCs and IgG4 increase in patients with T1DM, and they have a positive correlation, which deserves further research.
