Expression of DUOX2 in colon cancer and its effect on malignant behavior of SW620 and HCT116 cells

Abstract:

Objective

To detect the expression of dual oxidase 2 (DUOX2) in colon cancer tissues and investigate its effect on the malignant biological behavior of colon cancer cells.

Methods

Real-time quantitative reverse-transcription polymerase chain reaction (qRT-PCR) was used to detect the expression of DUOX2 in 97 pairs of colon cancer tissues and adjacent normal tissues which were collected from October 2018 to October 2019 at the Fourth Hospital of Hebei Medical University. siRNA was used to knock down the expression of DUOX2 in SW620 and HCT116 cells. MTS assay was used to determine the cell proliferation. Transwell and scratch assays were used to determine the migration and invasion of the cells.

Results

The relative expression of DUOX2 gene in colon cancer tissues was significantly higher than that in adjacent normal tissues [0.0265 (0.0061, 0.0869) vs 0.0087 (0.0017, 0.0385), P<0.05]. The expression of DUOX2 gene in colon cancer was correlated with gender, T stage, lymph node metastasis, and TNM stage (P<0.05). After knockdown of DUOX2 expression in SW620 and HCT116 cells, the migration and invasion ability decreased significantly (P<0.05), but the proliferation ability did not change significantly (P>0.05).

Conclusion

DUOX2 is highly expressed in colon cancer tissue. DUOX2 as an oncogene promotes the migration and invasion of colon cancer cells, suggesting that DUOX2 may be a biomarker for predicting metastasis and a potential target for treatment in colon cancer patients.
