目的

研究基于Toll样受体4(TLR4)/核因子κB(NF-κB)通路探究益气活血清热解毒方对动脉粥样硬化模型大鼠的干预效果。

方法

将60只大鼠随机分为正常组10只、其余50只大鼠建立动脉粥样硬化模型作为动脉粥样硬化组,喂养4周后每组随机抽样2只,取主动脉经HE染色光镜观察,以发现动脉粥样硬化斑块为建模成功,建模成功后将动脉粥样硬化组剩余48只大鼠分为模型组(9只)、辛伐他汀组(9只)、益气活血组(10只)、清热解毒组(10只)、益气活血清热解毒中药组(10只)5个亚组。建模成功后益气活血组、清热解毒组、益气活血清热解毒中药组分别给予药液9 ml/(kg·d)体重灌胃,辛伐他汀片组给予辛伐他汀药液5 ml/(kg·d)体重灌胃,正常组和模型组给予生理盐水9 ml/(kg·d),以上各组均连续灌胃8周。对比分析各组的血脂指标甘油三脂(TG)、血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白(HDL-C)、超敏C反应蛋白(Hs-CRP)、白介素-6(IL-6)水平及TLR4/NF-κB通路蛋白表达量。

结果

与正常组相比,模型组、辛伐他汀组、益气活血组、清热解毒组、益气活血清热解毒中药组TG、TC、LDL-C、Hs-CRP、IL-6水平升高,HDL-C水平降低,差异具有统计学意义(P<0.05);与模型组相比,辛伐他汀组、益气活血组、清热解毒组、益气活血清热解毒中药组TG、TC、LDL-C、Hs-CRP、IL-6水平降低,HDL-C水平升高,差异具有统计学意义(P<0.05);与辛伐他汀组、益气活血组、清热解毒组相比,益气活血清热解毒中药组TG、TC、LDL-C、Hs-CRP、IL-6水平降低,HDL-C水平升高,差异具有统计学意义(P<0.05)。与模型组相比,益气活血清热解毒中药组TLR4、NF-κB表达量降低(P=0.001)。

结论

益气活血清热解毒方可起到调节动脉粥样硬化大鼠血脂、缓解炎症症状的作用,其机制可能与抑制TLR4/NF-κB通路有关。

Objective

To explore the interventional effect of Yiqi Huoxue Qingre Jiedu (Qi-tonifying, blood circulation-activating, heat-clearing, and detoxification) recipe on atherosclerosis rats based on the TLR4/NF-κB pathway.

Methods

Sixty rats were randomly divided into either a normal group (10 rats) or an atherosclerosis group (50 rats; atherosclerosis was induced). After feeding for 4 weeks, two rats were randomly selected from each group, and the aorta was dissected and observed under a light microscope after HE staining to find atherosclerotic plaque. After successful modeling, the remaining 48 rats in the atherosclerosis group were divided into five groups: model group (9 rats), simvastatin group (9 rats), Qi-tonifying and blood circulation-activating group (10 rats), heat-clearing and detoxification group (10 rats), and Qi-tonifying, blood circulation-activating, heat-clearing, and detoxification group (10 rats). The Qi-tonifying and blood circulation-activating group, the heat-clearing and detoxification group, and the Qi-activating, blood circulation-activating, heat-clearing, and detoxification group were given the corresponding traditional Chinese medicine recipes at 9 ml/kg/d body weight, respectively; the simvastatin tablet group was given 5 mg/(kg·d) body weight of simvastatin solution, and the normal group and the model group were given 9 ml/kg/d of normal saline. All the treatments were given intragastrically for 8 consecutive weeks. The levels of TG, TC, LDL-C, HDL-C, hs-CRP, and IL-6 and the protein expression of TLR4 and NF-κB in different groups were compared and analyzed.

Results

Compared with the normal group, the levels of TG, TC, LDL-C, hs-CRP, and IL-6 were significantly increased in the model group, simvastatin group, Qi-tonifying and blood circulation-activating group, heat-clearing and detoxification group, and Qi-tonifying, blood circulation-activating, heat-clearing, and detoxification groups, while the level of HDL-C was significantly decreased (P<0.05). Compared with the model group, the levels of TG, TC, LDL-C, hs-CRP, and IL-6 were significantly decreased in the simvastatin group, Qi-tonifying and blood circulation-activating group, heat-clearing and detoxification group, and Qi-tonifying, blood circulation-activating, heat-clearing, and detoxification group, while the level of HDL-C was significantly increased (P<0.05). Compared with the simvastatin group, Qi-tonifying and blood circulation-activating group, and heat-clearing and detoxification group, the levels of TG, TC, LDL-C, hs-CRP, and IL-6 in the Qi-tonifying, blood circulation-activating,, heat-clearing, and detoxification group were significantly decreased, while the level of HDL-C was significantly increased (P<0.05). Compared with the model group, the protein expression of TLR4 and NF-κB was significantly reduced in the Qi-tonifying, blood circulation-activating,, heat-clearing, and detoxification group (P=0.001).

Conclusion

Yiqi Huoxue Qingre Jiedu recipe can regulate blood lipids and relieve inflammatory symptoms in atherosclerotic rats via mechanisms that may be related to the inhibition of the TLR4/NF-κB pathway.
